Mechanism. Copanlisib (BAY 80-6946) is the PI3Kα-binding POI WARHEAD of the best-validated PI3Kα PROTAC degrader D5 (DC50 0.05 nM); ATP-competitive orthosteric. This docking test scores POCKET ENGAGEMENT only — not degradation (which needs ternary-complex / E3 modeling) and not mutant-selectivity (the canine receptor is wild-type). A good pose is a necessary first step, not proof of a working degrader.
