Mechanism. Ipatasertib (GDC-0068) is an ATP-competitive AKT inhibitor. AKT1 is the surviving synthetic-lethal co-dependency of PIK3CA-mutant cells (DepMap 24Q4). This docking test scores pocket engagement on the verified AKT1 receptor; an approved AKT inhibitor binding AKT1 is expected — the value is establishing the dockable PI3Ka->AKT1 axis node for future novel-modality bets, not a surprising result.
